PD-168077 maleate is a selective dopamine D4 receptor agonist with a Ki of 9 nM. It exhibits high selectivity over other D2-like and D1-like receptor family members, as well as α1, α2, 5-HT1A, and 5-HT2A receptors. This compound shows intrinsic activity at the D4 receptor, inhibiting forskolin-stimulated cAMP accumulation or stimulating [3H]thymidine uptake in CHO cells expressing the human D4 receptor. In vivo, PD-168077 dose-dependently induces locomotion.
